Protoplast Culture of Flower Cabbage (Brassica oleracea L. Var . acephala DC . forma triocolor Hort .)

Abstract

|
The experiments were conducted to optimize several factors influencing protoplast culture from mesophyll of Brassica oleracea L. var acephala DC. forma tricolor Hort. `Eunbae´ and `Hongrin´. Preconditioned young leaves grown in a growth chamber for 3∼4 weeks were good for the source materials. Cell division and cell colony formation were the most vigourous when cultured with the cell density of 5 × 10⁴/㎖ in a 8p-KM medium containing 0.2M glucose and 0.3M mannitol with a pH 6.3. Cell colony formation was stimulated by adding some fresh 8p-KM medium without the 0.2M glucose and 0.3M mannitol after 7 days of culture.
